Simponi (generic name: golimumab) is a prescription medication classified as a tumor necrosis factor (TNF) blocker. It is designed to manage certain autoimmune conditions by inhibiting the activity of TNF-alpha, a substance in the body that causes inflammation. By targeting and neutralizing TNF-alpha, Simponi helps reduce inflammation and modulate abnormal immune responses. This medication is utilized in the treatment of several chronic inflammatory diseases:
  • Rheumatoid arthritis (RA)
  • Psoriatic arthritis (PsA)
  • Ankylosing spondylitis (AS)
  • Ulcerative colitis (UC).
By inhibiting TNF-alpha, Simponi plays a crucial role in controlling the inflammatory processes underlying these conditions, thereby improving patients’ quality of life and functional abilities.

Usual Dosage(s) and Mode(s) of Administration

Simponi is administered as a subcutaneous injection, allowing patients to self-administer the medication after proper training. The dosing schedule varies based on the specific condition being treated but generally involves once-monthly injections, providing a convenient treatment option for individuals managing chronic inflammatory diseases. The dosing regimen varies depending on the specific condition being treated:
  • Rheumatoid arthritis (RA): The recommended dose is 50 mg once a month, in combination with methotrexate.
  • Psoriatic arthritis (PsA): A 50 mg dose is administered once a month, which can be used alone or with methotrexate.
  • Ankylosing spondylitis (AS): Patients receive 50 mg once a month.
  • Ulcerative colitis (UC): The induction regimen starts with 200 mg at week 0, followed by 100 mg at week 2. For maintenance, 100 mg is administered every 4 weeks.
Patients are usually trained on the proper injection technique by a healthcare professional before self-administration. It’s important to rotate injection sites to avoid skin irritation. If a dose is missed, patients should inject the missed dose as soon as they remember and then continue with their regular monthly schedule. However, if the next dose is less than two weeks away, they should wait until the next scheduled dose.

Storage Guidelines

Simponi should be stored in a refrigerator at temperatures between 36°F to 46°F (2°C to 8°C). Do not freeze the medication. Keep the prefilled syringe or autoinjector in its original carton to protect it from light until the time of use. If needed, Simponi can be stored at room temperature, up to 77°F (25°C), for a maximum period of 30 days. Once stored at room temperature, it should not be returned to the refrigerator and must be discarded if not used within the 30-day period.
